What is it good for?

It is sold to soften expression lines, usually around the eyes and forehead, and is marketed as a topical alternative to an injected muscle relaxant. The two published studies that include it are tests of a whole patch containing several actives, not tests of this peptide.

What it is

SNAP-8 is a synthetic peptide of eight amino acids. On an ingredient label it appears under its INCI name, acetyl octapeptide-3, and occasionally as acetyl octapeptide-1.

It is sold to soften expression lines, and almost always positioned against an injected muscle relaxant — a topical alternative to a needle. The design story suppliers tell is that it mimics a fragment of SNAP-25, one of the proteins that docks vesicles at the nerve ending so that neurotransmitter can be released. The shorter peptide sold as Argireline carries the same story at six residues instead of eight, and SNAP-8 is marketed as the more potent of the two.

That is the claim. The record is smaller than the claim, and the interesting part is how much smaller.

The whole published record is two papers

PubMed holds 2 records naming this compound as of 2026-09-13:

  • Annals of Dermatology, 2024;36(4):215–224 (PMID 39082657) — a dissolving microneedle patch study, 24 healthy subjects, active patch on one eye and placebo patch on the other, assessed over 28 days.
  • Journal of Cosmetic Dermatology, 2020;19(2):328–337 (PMID 31134751) — bioactive peptides loaded on hyaluronic acid microneedle patches, a single-centre clinical study.

Both are genuine clinical work with human participants, published in peer-reviewed dermatology journals. Neither is a study of this peptide.

What the studies were actually comparing

In the 2024 paper the active patch is built from a hyaluronic acid backbone carrying acetyl octapeptide-3, L-ascorbic acid 2-glucoside and sodium cyclic lysophosphatidic acid. The placebo patch is hyaluronic acid and nothing else.

So the contrast is a patch with three actives against a patch with none. If the eye treated with the loaded patch does better, the result is consistent with the peptide working, with the vitamin C derivative working, with the lysophosphatidic acid working, with the microneedles themselves doing something, or with any combination of the four. The study was not designed to tell those apart, and it does not claim to. The 2020 paper has the same structure, announced in its own title: bioactive peptides, plural.

This is the check that costs one minute and is almost never done. A citation count tells you a compound appears in the literature. Reading what the compound was doing in each study — treatment, comparator, or one of several things held together — tells you whether any of it is evidence about the compound. Here, twice, the answer is no.

One further detail worth recording: Raphas Co. Ltd., the Korean company that manufactures the microneedle patches, appears as an author affiliation on both papers. That is ordinary in cosmetic science, and it is not a reason to discard either result. It does mean that the entire published literature on this ingredient comes from work connected to one patch maker.

The 92 that are not there

Search PubMed for the loose phrase acetyl octapeptide and it returns 92 records, a number that would make this compound look reasonably well studied.

The first fifteen of those by date include lentiviral vector promoters for a sulfatase gene, multi-O-GalNAcylated MUC5AC tandem repeats, the Pax5 C-terminal domain in B-cell development, peptaibol total synthesis, and angiotensin-converting-enzyme inhibitors. They match because they contain the word acetyl and the word octapeptide, in unrelated senses.

Search the ingredient names and the answer is two. The gap between 92 and 2 is not a subtlety of search syntax; it is the difference between a compound with a literature and a compound with two papers, and it resolves by reading the titles. This library has now found the same error class on several compounds, and it runs in both directions — a count that is too high because the search is loose, and a count that is too low because the trial was filed under a name the market does not use.

There is no approved medicine containing this peptide, and no application for one. Drugs@FDA returned nothing on 2026-09-13.

How to read this entry

The honest summary is narrow. A microneedle patch containing this peptide and other actives improved eye wrinkles against an empty patch in a 24-person study, with no reported adverse effects. Everything beyond that sentence — that the peptide is responsible, that it works in a cream rather than on a needle, that eight residues beat six, that it does anything to a nerve — is either unmeasured or untested.

Searching the actual ingredient names returns two. Counting a search result without reading it is the single most common error in peptide marketing.

Does it work like an injected muscle relaxant?

Nothing in its published record tests that. The comparison is a marketing frame built on the peptide's stated design, and the two clinical studies measure wrinkle appearance, water loss and elasticity — not nerve signalling or muscle contraction. A topical peptide and an injected neurotoxin also differ in the more basic respect that one has to cross the outer layer of skin to reach anything, which is why both published studies deliver it on a microneedle patch rather than in a cream.

Has anyone reported harm from it?

No safety signal appears in the two published studies, both of which report no adverse effects. With a total of roughly 24 to 40 participants across them and no registered trial, that is an absence of findings rather than a safety record.

What would change the picture?

One study comparing the same patch with and without this single peptide in it. That design is straightforward, inexpensive, and would answer the question the existing two papers cannot. Its absence after more than fifteen years on the market is itself informative.
